چکیده

the brain and spinal cord have a limited capacity for self-repair under damaged conditions. one of the best options to overcome these limitations involves the use of phytochemicals as potential therapeutic agents. in this study, we have aimed to investigate the effects of di-(2-ethylhexyl) phthalate (dehp) on hippocampus-derived neural stem cells (nscs) proliferation to search phytochemical candidates for possible treatment of neurological diseases using endogenous capacity. in this experimental study, neonatal rat hippocampus-derived nscs were cultured and treated with various concentrations of dehp (0, 100, 200, 400 and 600 µm) and cirsium vulgare (c. vulgare) hydroethanolic extract (0, 200, 400, 600, 800 and 1000 µg/ml) for 48 hours under in vitro conditions. cell proliferation rates and quantitative sox2 gene expression were evaluated using mtt assay and real-time reverse transcription polymerase chain reaction (rt-pcr). we observed the highest average growth rate in the 400 µm dehp and 800 µg/ml c. vulgare extract treated groups. sox2 expression in the dehp-treated nscs significantly increased compared to the control group. gas chromatography/mass spectrometry (gc/ms) results demonstrated that the active ingredients that naturally occurred in the c. vulgare hydroethanolic extract were 2-ethyl-1-hexanamine, n-heptacosane, 1-cyclopentanecarboxylic acid, 1-heptadecanamine, 2,6-octadien-1-ol,2,6,10,14,18,22-tetracosahexaene, and dehp. dehp profoundly stimulated nscs proliferation through sox2 gene overexpression. these outcomes could be an invaluable opportunity for further use of the c. vulgure phytochemicals for prevention and/or treatment of neurological diseases via phytochemical mediated-proliferation of endogenous adult nscs.

Di-(2-ethylhexyl) Phthalate-Induced Hippocampus-Derived Neural Stem Cells Proliferation

The brain and spinal cord have a limited capacity for self-repair under damaged conditions. One of the best options to overcome these limitations involves the use of phytochemicals as potential therapeutic agents. In this study, we have aimed to investigate the effects of di-(2-ethylhexyl) phthalate (DEHP) on hippocampus-derived neural stem cells (NSCs) proliferation to search phytochemical can...

Alpha-Tocopherol increases the proliferation of induced pluripotent stem cell derived neural progenitor cells

In addition to its antioxidant effect, Vitamin E or α–tocopherol is suggested to enhance remyelination in the animal model of non-inflammatory demyelination. In this study, the possible proliferative effect of vitamin E on human- induced pluripotent stem cell-derived neural progenitors (hiPS-NPs) and the underlying mechanisms were investigated in vitro. NPs were induced from iPS cells via 3 ste...

Effect of Di(2-ethylhexyl)phthalate on Helicobacter pylori-Induced Apoptosis in AGS Cells

Plastic products are wildly used in human life. Di(2-ethylhexyl)phthalate (DEHP) is an essential additive in plastic manufacturing and is used as plasticizer for many products including plastic food packaging. DEHP is a teratogenic compound and can cause potent reproductive toxicity. DEHP can also cause liver damage, peroxisome proliferation, and carcinogenesis. DEHP is also strongly associated...

Teratogenicity of di(2-ethylhexyl) phthalate (DEHP) and di-n-butyl phthalate (DBP) in mice.

Di(2-ethylhexyl) phthalate (DEHP) and di-n-butyl phthalate (DBP) were mixed with diet at graded levels of 0.05, 0.1, 0.2. 0.4 and 1.0 wt-% and given to pregnant ICR mice throughout gestation. Maternal weight gain was suppressed and fetal resorption increased at 0.2, 0.4 and 1.0% levels of DEHP and 1.0% level of DBP. All the implanted ova died early in rats fed 0.4 and 1.0% levels of DEHP. Exter...

Effects of di-isononyl phthalate, di-2-ethylhexyl phthalate, and clofibrate in cynomolgus monkeys.

The effects of the peroxisome proliferators di-isononyl phthalate (DINP) and di-2-ethylhexyl phthalate (DEHP) were evaluated in young adult male cynomolgus monkeys after 14 days of treatment, with emphasis on detecting hepatic and other effects seen in rats and mice after treatment with high doses of phthalates. Groups of 4 monkeys received DINP (500 mg/kg/day), DEHP (500 mg/kg/day), or vehicle...
